Universalness_h (BBa_K5086009) is a single-stranded DNA sequence with 30 nucleotides that is complementary to the 3' end sequences of hsa-mir-99b_DDSD, hsa-mir-200c_DDSD, hsa-mir-21_DDSD, and hsa-mir-141_DDSD probes ' end of the sequence is complementary to the 5' end of the hsa-mir-99b_Rep, hsa-mir-200c_Rep, hsa-mir-21_Rep, and hsa-mir-141_Rep probes, and therefore can specifically bind to their 5 ' end and is an invariant part of the pluggable probe sequence. In the reaction DNA polymerase polymerizes DNA in the 5'-3' direction starting from the 3' end of the target, and during DNA polymerization by Bst DNA polymerase, the Universalness_h strand is displaced and released by the newly synthesized daughter strand. Through this process, the target signal is converted into the Universalness_h strand (the significance of this strand replacement step: to facilitate the realization of the subsequent self-enhancement, if there is no replacement of the Universalness_h strand this reaction can not be synthesized by the DDSD probe shelf). The replaced Universalness_h chain will be further combined with hsa-mir-99b_DDSD, hsa-mir-200c_DDSD, hsa-mir-21_DDSD, and hsa-mir-141_DDSD probes for subsequent chain replacement reactions to generate fluorescence reports.
